產品簡介：
本產品適用于SDS-PAGE(SDS變性聚丙烯酰胺凝膠電泳)時作蛋白質上樣用。其主要成份為SDS，β-巰基乙醇，溴酚藍，緩沖鹽溶液等。 SDS可與蛋白質結合使蛋白質－SDS復合物上帶有大量的負電荷，這時蛋白質本身的電荷完全被SDS掩蓋，消除了各種蛋白質本身電荷的差異，SDS還可以斷開分子內和分子間的氫鍵，破壞蛋白質分子的二級和三級結構；β-巰基乙醇可以斷開半胱氨酸殘基之間的二硫鍵，破壞蛋白質的四級結構，消除了蛋白結構之間的差異。最終無電荷及結構上差異的蛋白（亞單位），電泳速度只是與其分子量大小有關；溴酚藍用作電泳時的指示劑，可大概指示電泳結束的時間。
使用說明：
1. 請按每30μL蛋白樣品加入10μL上樣緩沖液的比例（4倍稀釋）來使用。如果蛋白濃度過高，可用雙蒸水稀釋樣品。
2. 混勻后，100℃水浴加熱3-5分鐘，使蛋白變性。
3. 冷卻到室溫后，10000-14000rpm離心2-5分鐘，取上清直接上樣電泳即可。
注意事項：
1. 聚丙烯酰胺凝膠濃度為8%時溴酚藍指示條帶的位置大概在30kd左右；膠濃度12%時，約在20kd左右；膠濃度15%時，大概在10kd。請根據自己目標條帶來判斷結果電泳時間。
2. 本試劑因含β-巰基乙醇，有一定的毒性，為了您的安全和健康，請穿實驗服并戴一次性手套操作。
3. 蛋白上樣緩沖液含有溴酚藍指示劑，PH值受保存溫度影響，在低溫凍存狀態下，溶液可能會呈現深棕色，不影響產品使用。
